Jesus's declaration to Nicodemus that 'except a man be born of water and of the Spirit, he cannot enter into the kingdom of God' has generated sustained debate about the referent of 'water.' The three major interpretive options are: (1) baptismal water (patristic and Catholic tradition), (2) physical birth (water of the womb, contrasted with Spirit-birth in verse 6), or (3) the word of God as the cleansing water (Ephesians 5:26; Titus 3:5). The context of John 3 — where Nicodemus is a Jewish teacher who should understand these things (verse 10) — suggests an OT background in Ezekiel 36:25-27's promise of water-cleansing and Spirit-indwelling in the new covenant. Titus 3:5's 'washing of regeneration, and renewing of the Holy Ghost' combines water and Spirit in a way that suggests their inseparability rather than requiring a choice between them.
